During the surveys of cabbage diseases in the years 2009-2012, mature plants of both white cabbage (B. o. var. capitata f. alba) and red cabbage (B. o. var. capitata f. rubra) with yellowing and detaching of lower leaves, watersoaking, browning and softening of leaf petioles and head outer parts, formation of white cottony mycelium and numerous large and black sclerotia on diseased parts, characteristics of Sclerotinia head rot of cabbage, was observed (Fig. 1 in Persian section). Infected heads or petiols and sclerotia were collected, surface sterilized and grown on PDA medium. Isolated fungi were purified by hyphal tip culture method and were kept in PCA slants for further studies. Apothecia were produced from sclerotia based on the method of Mylchreest and Wheeler (1987). Based on morphological characters of isolated fungi, Sclerotinia sclerotiorum (Lib.) de Bary was identified (Purdy, 1979; Saharan and Mehta, 2008). Pathogenicity tests were performed both on detached leaves and on container-grown healthy, 3 months old cabbage stems. Healthy cabbage leaves were washed under tap water, and then surface sterilized with 10% ethanol and inoculated at the center with a 5 mm2 PDA plug from 5 days old mycelial cultures. The inoculated leaves were kept in plastic containers equipped with wet filter paper and incubated at room temperature for 10 days. Cabbage stems were first surface sterilized and then inoculated as leaf assays without any wounding. Plants were incubated in a growth chamber and maintained at 25°C, 95% relative humidity on a 12-h fluorescent light/dark conditions for 7 days. Leaves and stems only inoculated with PDA plugs served as controls. Inoculated leaves and stems showed brown, water soaked and softened lesions several centimeters around the inoculated area (Fig. 2 in Persian section). No symptoms were observed on control plants. Inoculated fungi reisolated from symptomatic parts, so Kochs postulates were completed. This is the first report of cabbage head rot on white and red cabbages caused by Sclerotinia sclerotiorum from Iran. in Iran.}, keywords = {}, title_fa = {اولین گزارش از وقوع بیماری پوسیدگی اسکلروتینیایی کلم پیچ در ایران}, abstract_fa = {  گیاه کلم پیچ با نام علمی Brassica oleracea var. capitata یکی از سبزیجات مهم اقتصادی و با سطح وسیع کشت در شهرستان ارومیه از استان آذربایجان غربی می‌باشد. طی بررسی بیماری‌های کلم در فاصله سال‌های 1391-1389، گیاهان بالغ کلم‌های سفید (B. o. var. capitata f. alba) و کلم‌های قرمز (B. o. var. capitata f. rubra) (شکل 1) با نشانه‌های زرد شدن و ریزش برگ‌های بیرونی بوته‌ها، آبسوختگی، قهوه‌ای شدن و نرم شدن قاعده دمبرگ‌ها و برگ‌های بیرونی سر کلم، تشکیل میسیلیوم سفید رنگ پنبه‌ای و تعداد زیادی سختینه‌های سیاه رنگ و بزرگ، که از مشخصات پوسیدگی اسکلروتینیایی کلم است، مشاهده شدند. بافت‌های آلوده به همراه سختینه‌ها از مزارع مختلف جمع‌آوری شدند. بافت‌های آلوده و سختینه‌ها ابتدا توسط محلول هیپوکلریت سدیم 5/0 درصد ضدعفونی سطحی شدند و بعد از 3 بار شستشو با آب مقطر سترون و خشک نمودن بین لایه‌های کاغذ صافی سترون، قطعات کوچکی از آنها برش داده شد و در تشتک‌های پتری حاوی محیط کشت PDA کشت شدند. قارچ‌های رشد کرده با روش کشت نوک ریسه خالص‌سازی شدند و داخل لوله‌های آزمایش حاوی محیط کشت PCA برای مطالعات بعدی نگهداری شدند. برای تولید آپوتسیوم‌ها از سختینه‌ها بر اساس روش میلچریست و ویلر(Mylchreest and Wheeler (1987)) عمل گردید. بر اساس مشخصات مورفولوژیکی قارچ‌های جدا شده، گونه Sclerotinia sclerotiorum (Lib.) de Bary.  تشخیص داده شد (پوردی، 1979؛ سهاران و مهتا، 2008). آزمون‌های بیماری‌زایی روی برگ‌ها و نیز ساقه‌های گیاهان سالم کلم سه ماهه، کشت شده در گلدان، انجام شد. برگ‌های سالم کلم پس از شستشو با آب و ضدعفونی سطحی با اتانول 10 درصد با حلقه‌های 5 میلی‌متری از کشت‌های فعال و 5 روزه قارچ روی محیط کشتPDA  تلقیح شدند. برگ‌های تلقیح شده در ظروف پلاستیکی که حاوی کاغذ صافی مرطوب بودند در دمای اتاق به مدت 10 روز نگهداری شدند. ساقه‌های کلم هم ابتدا ضدعفونی سطحی شدند و سپس همانند برگ‌ها و بدون ایجاد هیچ‌گونه زخم، تلقیح شدند. گیاهان تلقیح شده در یک محفظه رشدی در دمای 25 درجه سلسیوس و رطوبت نسبی 95 درصدو دوره نوری/تاریکی 12 ساعته زیر نور فلورسنت به مدت 7 روز نگهداری شدند. از حلقه‌های آگاری فاقد قارچ برای تلقیح گیاهان شاهد استفاده شد. برگ‌ها و ساقه‌های تلقیح شده علائم قهوه‌ای شدن، آبسوختگی و نرم شدگی در اطراف محل تلقیح را بعد از 7-3 روز نشان دادند (شکل 2). هیچ علائمی در گیاهان شاهد ایجاد نشد. قارچ تلقیح شده دوباره از محل آلودگی جداسازی شد و بدین ترتیب اصول کخ اثبات شد. این اولین گزارش از آلودگی گیاهان کلم سفید و قرمز به قارچ Sclerotinia sclerotiorum از ایران است.}, keywords_fa = {}, url = {https://ijpp.areeo.ac.ir/article_15435.html}, eprint = {https://ijpp.areeo.ac.ir/article_15435_17933d02f03f85eaa0e7be973ce123e5.pdf} }
