The causal pathogen is Erwiniaamylovora, species in the family Enterobacteriaceae. Pears are the most susceptible, but apples, quinces and some other rosaceous plants are also vulnerable. In Iran, the disease has been reported for the first time in Baraghan in Alborz province (Zakeri & Sharifnabi 1991). Fire blight produces several different types of symptoms like blossom blight, necrosis of fruits and leaf tissues, and dark sunken cankers on stems and exudation of droplets of bacterial ooze from the affected plant tissues. Typical symptoms of fire blight were observed on quince (Cydonia oblonga) trees, in Shahrbabak in Kerman province in 2013. Dieback of terminal shoots and dark necrotic lesions on immature fruits were among the major symptoms observed. Samples containing regions of dead and healthy tissues were washed, macerated in drops of sterile distilled water and bacteria were isolated on sucrose nutrient agar (NAS) medium. After 72 h of incubation at 27°C, single whitish and round colonies were purified by sub-culturing on eosine methylene blue agar (EMB agar). Results of morphological and biochemical tests, demonstrated that the bacterium belongs to Enterobacteriaceae (Schaad et al. 2001). Pathogenicity was tested by injecting healthy young quince shoots with a 10-μl bacterial suspension of 107CFU/ml. Controls were injected with sterile distilled water. Shoots were kept at 27°C and 80 to 100% relative humidity. Dark brown-to-black lesions were observed five days after inoculation on the inoculated shoots. The bacterium was reisolated from the lesions formed on the inoculated shoots. No lesions were observed on the controls. All of the twenty strains used in tests for identification, produced white colonies on NAS, were gram-negative, induced a hypersensitive reaction in tobacco (Nicotiana tabacum L. ‘White Burley’) leaves, did not produce a fluorescent pigment on medium B of King and did not grow at 37°C. On the basis of the symptoms, colony morphology and biochemical features, the pathogen was identified as E. amylovora. To our knowledge, this is the first report on the incidence of fire blight on quince in Kerman province.}, keywords = {}, title_fa = {وقوع بیماری آتشک به با عامل Erwinia amylovora در استان کرمان}, abstract_fa = {  *بخشی از پایان‌نامه کارشناسی‌ارشد نگارنده اول، ارائه شده به دانشکده کشاورزی دانشگاه شهید باهنر کرمان ** مسئول مکاتبات، پست الکترونیکی: Pkhodaygan@vru.ac.ir 1 - گروه گیاهپزشکی دانشگاه شید باهنر کرمان 2- گروه گیاهپزشکی دانشگاه ولی عصر رفسنجان   بیماری آتشک یکی از مخرب‌ترین بیماری‌های باکتریایی درختان هسته‌دار و دانه‌دار می‌باشد. عامل بیماری، Erwinia amylovora از اعضای خانواده Enterobacteriaceae است. از میان درختان میوه، گلابی حساس‌ترین میزبان و درختانی نظیر سیب، به و برخی دیگر از اعضای خانواده روزاسه نیز به این بیماری حساس هستند. در ایران آتشک برای نخستین بار از برغان کرج گزارش گردید (Zakeri & Sharifnabi 1991). این بیماری سبب بروز علائم متعددی نظیر بلایت شکوفه‌ها، نکروز و تراوش صمغ شیری رنگ باکتری از اندام‌های آلوده می‌گردد. در سال 1391 علائم این بیماری شامل سرخشکیدگی شدید و سیاه‌شدگی میوه‌های نارس به (Cydonia oblonga)، در مناطقی از شهرستان شهربابک در استان کرمان مشاهده گردید. نمونه‌هایی در فصل بهار و پائیز از باغات مختلف به تهیه و برای جداسازی بیمارگر به آزمایشگاه منتقل گردید. نمونه‌هایی از بافت آلوده تهیه شده و روی محیط کشت اگار غذایی حاوی نیم درصد ساکاروز کشت داده شد. پس از 72 ساعت نگهداری در دمای 27 درجه سلسیوس کلنی‌های سفید و لعاب‌دار در حال رشد بر سطح محیط کشت جدا و روی محیط EMB مخطط گردید. به منظور شناسایی جدایه‌ها از روش‌های مرفولوژیک و بیوشیمیایی استفاده گردید( Schaad et al. 2001). برای بررسی توان بیماری‌زایی جدایه‌ها، سرشاخه‌های جوان به با سوسپانسیونی حاوی 107 کلنی در هر میلی‌لیتر، مایه‌زنی شده و در شرایط گلخانه با رطوبت نسبی 80 تا 100 درصد و دمای 27 درجه سلسیوس نگهداری گردید. به عنوان کنترل منفی از آب مقطر استریل برای مایه‌زنی سرشاخه‌ها استفاده گردید. پنج روز پس از مایه‌زنی علائم بیماری بصورت زخم‌های تیره در سرشاخه‌ها، ایجاد گردید. در سرشاخه‌هایی که با آب مایه‌زنی شده بودند علائمی دیده نشد و از بافت‌های آلوده باکتری بیمارگر مجددا جداسازی گردید. بیست جدایه‌ باکتری در این تحقیق مورد ارزیابی قرار گرفتند. این جدایه‌ها روی محیط کشت NAS کلنی‌های سفیدی ایجاد نموده، گرم‌منفی، بی‌هوازی اختیاری بوده و سبب القا واکنش فوق‌حساسیت در توتون شدند. جدایه‌ها اکسیداز منفی بوده و توانایی تولید رنگ فلورسنت روی محیط کشت Kings B و رشد در دمای 37 درجه سلسیوس را نداشتند. براساس ویژگی‌های فنوتیپی و بیوشیمیایی تعیین شده و آزمون بیماری‌زایی، جدایه‌ها به عنوان باکتری E. amylovora تشخیص داده شدند. بر اساس اطلاعات ما این نخستین گزارش از وجود بیماری آتشک به، در استان کرمان است.}, keywords_fa = {}, url = {https://ijpp.areeo.ac.ir/article_15781.html}, eprint = {https://ijpp.areeo.ac.ir/article_15781_bd31afef0f738bde578911acca662f49.pdf} }
