This pathogen causes yield loss in field and tuber rot in storage conditions. Genetic diversity and pathogenicity of C. coccodes was unknown in Iran. In this study, C. coccodes was isolated from potato plants with black dot symptoms from Ardebil, Isfahan and Hamadan provinces. Amongst 108 isolates, 48 isolates were studied for vegetative compatibility groups and aggressiveness. Ten VCGs including four multi-member groups (IRN-VCG1, IRN-VCG2, IRN-VCG3 and IRN-VCG4) as well as  six single-member groups were detected. Isolates of Ardebil were grouped in IRN-VCG1and IRN-VCG2 with 50 and 12.5% frequency and other isolates were single-member. Three groups of IRN-VCG1 (55%), IRN-VCG2 (13%) and IRN-VCG3 (13%) were constituted for Hamadan province isolates. Isfahan isolates were grouped as IRN-VCG1, IRN-VCG2 and IRN-VCG4 at 25, 19 and 12 percentage respectively, and 44% of isolates were not assigned to any of the four VCGs. The aggressiveness of the isolates were investigated using potato tuber decay assessment and root inoculation method. Most of isolates were able to decay potato tubers, while all isolates colonized the roots and were considered as aggressive isolates. Comparison of root and tuber inoculation methods showed no relationship between tuber rot severity and microsclerotia spreading in roots and stems. There was no correlation between VCGs and virulence while some correlation was detected for VCGs and geographical origin of the isolates. This study demonstrated that there is a significant VCG diversity in C. coccodes.}, keywords = {Genetic diversity,Potato early dying,nit mutant,disease diversity,Colletotrichum coccodes}, title_fa = {بررسی گروه‌های سازگاری رویشی و بیماری‌زایی جدایه‌های Colletotrichum coccodes عامل خال سیاه سیب‌زمینی در ایران *}, abstract_fa = {بیماری خال سیاه سیب‌زمینی توسط Colletotrichum coccodes ایجاد می‌شود و در اکثر مناطق کشت سیب‌زمینی ایران پراکنده است. عامل بیماری یکی از عوامل مهم کاهش عملکرد در مزرعه است و باعث پوسیدگی غده‌ها و افت وزن محصول در انبار می‌شود. وضعیت تنوع ژنتیکی و توان بیماری‌زایی این قارچ در ایران مشخص نبود. برای این منظور از مناطق عمده سیب‌زمینی کاری استان‌های اردبیل‌‍، اصفهان و همدان اقدام به نمونه‌برداری از گیاهان با علایم بیماری شد. از 108 جدایه به دست آمده 48 جدایه با توجه به مناطق نمونه برداری انتخاب شدند. بر پایه واکنش‌های سازگاری رویشی در مجموع چهار گروه چند عضوی IRN-VCG1، IRN-VCG2، IRN-VCG3 و IRN-VCG4 به ترتیب با فراوانی 86/42، 29/14، 16/4 و 16/4 درصد  و شش گروه تک عضوی تعیین شدند. جدایه‌های اردبیل در گروه‌های IRN-VCG1 و IRN-VCG2 با فراوانی 50 و 5/12 درصد قرار گرفتند و بقیه جدایه‌ها گروه‌های تک عضوی را تشکیل دادند. سه گروه IRN-VCG1، IRN-VCG2 و IRN-VCG3 به ترتیب با فراوانی 55، 13 و 13 درصد جدایه‌های همدان را در بر گرفتند و باقی جدایه‌ها در گروه‌های تک عضوی جای داده شدند. جدایه‌های استان اصفهان به سه گروه IRN-VCG1، IRN-VCG2 و  IRN-VCG4با فراوانی 25، 19 و12 درصد تعلق داشتند و بقیه جزو گروه‌های تک عضوی بودند. با استفاده از دو روش تلقیح اینوکلوم قارچ به غده‌های سیب‌زمینی و آلوده‌سازی ریشه، بیماری‌زایی جدایه‌ها بررسی شد. به جز پنج جدایه بقیه جدایه‌ها قادر به پوسانیدن غده‌های سیب‌زمینی بودند و همه جدایه‌ها توانستند در ریشه و ساقه زیرزمینی ایجاد بیماری کنند. مقایسه داده‌های مربوط به شدت بیماری‌زایی نشان داد شدت پوسانندگی غده‌ها و گسترش میکرواسکلروت‌های جدایه‌ها در اندام‌های زیرزمینی مستقل از هم هستند. هم‌چنین بین گروه‌های سازگاری رویشی و قدرت بیماری‌زایی جدایه‌ها هم‌بستگی وجود نداشت ولی بین این گروه‌ها و مناطق جغرافیایی جدایه‌ها هم‌بستگی اندکی دیده شد. پژوهش حاضر نشانگر تنوع قابل توجه گروه‌های سازگاری رویشی در این گونه است.}, keywords_fa = {تنوع ژنتیکی,مرگ زودرس سیب‌زمینی,جهش یافته nit,شدت بیماری,Colletotrichum coccodes}, url = {https://ijpp.areeo.ac.ir/article_15809.html}, eprint = {https://ijpp.areeo.ac.ir/article_15809_029e71c538dbfa2eddb6e5c129a872a0.pdf} }
