Using resistant cultivars is still the most effective method of management of this disease. However, for screening cultivars for resistance, knowledge of genetic diversity of a fungal population is necessary. In this study, genetic diversity of the pathogen in Tehran, Hamedan and Ardebil provinces was studied. Seventy isolates of F. oxysporum were isolated from infected plants. Pathogenic variability of the isolates in terms of degree of  wilting and root-rotting was tested and their genetic diversity was studied using seven RAPD primers. Pathogenicity tests were based on completely randomized design (CRD) with four replicates. The result of pathogenicity tests showed that isolates caused potato wilt had lower root-rot potential and vice versa. Cluster analysis of RAPD data using UPGMA method and Dice’s coefficient distinguished 8 main groups at 71% similarity level. Also, 18 molecular phenotypes were detected among isolates. The low number of distinguished groups observed among the populations could suggest that these isolates have probably originated from a common ancestor and sexual reproduction has played a minor role in genetic variability of the fungus in its life cycle. Also no apparent correlation was found between RAPD groups and geographical region and it suggests that they are independent from each other.}, keywords = {}, title_fa = {بررسی تنوع ژنتیکی Fusarium oxysporum عامل پژمردگی فوزاریومی سیب زمینی با استفاده از آزمون‌های بیماری‌زایی و نشانگرهای RAPD}, abstract_fa = {                گونه‌ی  Fusarium oxysporum عامل بیماری پژمردگی فوزاریومی سیب زمینی در اغلب مزارع سیب زمینی کاری ایران شایع بوده و خسارت زیادی می‌زند. استفاده از ارقام مقاوم یکی از مؤثرترین روش‌های مدیریت این بیماری محسوب می‌شود، اما به منظور انتخاب و توسعه ارقام مقاوم آگاهی از تنوع ژنتیکی جمعیت‌های این قارچ ضروری است. در این بررسی تنوع ژنتیکی جمعیت این بیمارگر در استان‌های تهران، همدان و اردبیل بررسی گردید. بدین منظور70 جدایه قارچ F. oxysporum به دست آمده از استان‌های مذکور انتخاب و پس از انجام آزمون بیماری‌زایی به دو روش آزمون بیماری‌زایی توانایی پوسانندگی ریشه و انسداد آوندی، با استفاده از هفت آغازگر RAPD تنوع ژنتیکی آن‌ها بررسی شد. آزمو‌ن‌های بیماری‌زایی در قالب طرح کاملاً تصادفی با چهار تکرار انجام شد. نتایج حاصل از آزمون بیماری‌زایی جدایه‌ها نشان داد جدایه‌هایی که در سیب زمینی پژمردگی ایجاد می‌نمودند از توان پوسانندگی کمی برخوردار بوده و بالعکس. تجزیه خوشه‌ای داده‌های RAPD با استفاده از روش UPGMA و ضریب تشابه Dice جدایه‌ها را در سطح شباهت 71 درصد در هشت گروه ژنوتیپی قرار داد. همچنین جدایه‌ها از لحاظ فنوتیپ مولکولی (هاپلوتیپ) به 18 گروه تقسیم شدند. تعداد اندک گروه‌بندی نشان دهنده تنوع ژنتیکی کم این قارچ در مناطق مورد بررسی می‌باشد و نشان می‌دهد که جدایه‌ها از یک منبع اجدادی منشأ گرفته‌اند که یکی از علل این امر را می‌توان به نقش ضعیف تولید مثل جنسی در این قارچ نسبت داد. همچنین تجزیه خوشه‌ای داده‌های RAPD جدایه‌ها را بدون توجه به منشأ جغرافیایی در گروه‌های ژنوتیپی مختلف قرار داد. بدین ترتیب بین تنوع ژنتیکی و منشأ جغرافیایی جدایه‌ها ارتباطی مشاهده نگردید.}, keywords_fa = {بیماری‌زایی,پوسیدگی,پژمردگی فوزاریومی,فنوتیپ مولکولی,سیب زمینی}, url = {https://ijpp.areeo.ac.ir/article_17607.html}, eprint = {} }
