The disease symptoms of the cultivated agaric include brown blotch, brown pit spots, rotting, weeping and drippy gill. Pseudomonas tolaasii has been cited as the causal agent of bacterial blotch, but much controversy exists regarding the involvement of this bacterium alone or blotch may be caused by more than one organism. In 2011, the diseased samples of mushroom were collected from Khorasan Razavi, Alborz, Tehran, Semnan, Ghazvin and Golestan provinces. Strains which were pathogenic on the cap of mushroom (pileus) were selected for further analysis. Biochemical, physiological and nutritional tests together with amplification of 1Kb region of 16S rDNA using Ps forward and reverse primers showed that all strains belong to Pseudomonas tolaasii. The isolated strains displayed different degrees of pathogenicity on mushroom. The presence of different virulence factors were evaluated among the strains. The results indicated that those strains which showed enhanced pyoverdin production had higher pathogenicity on agaric caps. There was no correlation between the existence of other virulence factors such as biofilm formation, motility, or PQS production and pathogenicity. Our results indicated that in P. tolaasii similar to other Pseudomonads, PQS exists but the actual role of this signal is not so clear. PQS dependent quorum sensing in P. tolaasii could not affect pathogenicity of this bacterium, thus most of the virulence determinants in this bacterium should be PQS independent.}, keywords = {Pseudomonas,Agaricus bisporus,PQS,Iran}, title_fa = {شناسایی و بررسی برخی فاکتورهای بیماریزایی جدایه‌های Pseudomonas tolaasii جدا شده از قارچ خوراکی تکمه‌ای سفید در ایران}, abstract_fa = {بیماری لکه قهوه‌ای قارچ خوراکی تکمه‌ای سفید (Agaricus bisporus) در سال‌های اخیر به عنوان یک بیماری شایع در سالن‌های پرورش قارچ ایران، ایجاد مشکل نموده است. علائم بیماری به صورت، لکه‌های کوچک قهوه‌ای، سوختگی قهوه‌ای و کلاهک‌های پوسیده که حالت لزج و چسبناکی دارند مشاهده می‌شود. باکتری Pseudomonas tolaasii به عنوان عامل اصلی ایجاد لکه قهوه‌ای مطرح می‌باشد. اما در منابع همیشه بیش از یک میکروارگانیسم به عنوان عامل لکه قهوه‌ای در نظر گرفته می‌شود. نمونه برداری از کلاهک قارچ‌های دارای علائم مشکوک از استان‌های البرز، خراسان رضوی، تهران، سمنان و قزوین در سال 1390 صورت پذیرفت. آزمونهای مختلف بیوشیمیایی، فیزیولوژیکی، تغذیه‌ای و تکثیر ناحیه یک کیلو بازی از rDNA S16 با استفاده از آغازگرهای رفت و برگشت (Ps-F, Ps-R) مشخص نمود که جدایه‌ متعلق به جنس Pseudomonas و گونهPseudomonas tolaasii هستند. آزمون اثبات بیماریزایی جدایه‌ها بر روی کلاهک قارچ تکمه‌ای درجات مختلفی از شدت بیماریزایی را در بین جدایه‌ها نشان داد. با بررسی برخی از فاکتورهای بیماریزایی مشخص گردید، جدایه‌هایی که بیشترین توان بیماریزایی را دارند از قدرت تولید سیدروفور پایووردین بالاتری برخوردار می‌باشند. در هیچ یک از جدایه‌ها، ارتباط مستقیمی بین تولید سایر فاکتورهای بیماریزایی باکتری‌ها از جمله تولید بیوفیلم، تولید تولاسین، تحرک و تولید سیگنال (PQS) Pseudomonas quinolone signal مشاهده نشد. نتایج این تحقیق نشان داد که در باکتری Pseudomonas tolaasii، همانند اغلب سودوموناس‌ها، PQS وجود دارد اما نقش واقعی آن مشخص نیست.}, keywords_fa = {سودوموناس,قارچ خوراکی,PQS,ایران}, url = {https://ijpp.areeo.ac.ir/article_19618.html}, eprint = {https://ijpp.areeo.ac.ir/article_19618_6ff24cd708f8e7196c3b5632ca993805.pdf} }
