تأثیرعلفکش گلیفوسیت در بیان ژنهای مرتبط با بیماریزایی و القای مقاومت در گیاهان سیبزمینی تراریخت تیمارشده با دو سویه بیمارگرهای سیبزمینی
هدف اصلی، بررسی تأثیر گلیفوسیت در بروز القای مقاومت نسبت به باکتریهای بیماریزای سیبزمینی بود. بدین منظور، از علفکش گلیفوسیت در غلطت بهینه 8/1 میلیگرم در لیتر بر رقم سیبزمینی تراریخت جهت القای مقاومت به دو سویه از باکتریهای بیمارگر سیبزمینی (سویه 21A از باکتری Pectobacterium atrosepticum و سویه ENA49 از باکتری Dickeya dadantii) استفاده شد. آنالیز PCR نمونه های cDNA جداشده و سنتزشده از گیاهان تراریخت نشان داد که ژنهای aroA و ژنهای پاسخ دفاعی سیب زمینی به میزان بیشتری بیان شدند. در اثر آلودگی برگهای سیبزمینی با باکتریهای بیمارگر، و تیمار آنها با گلیفوسیت، یک سطح بالایی از بیان ژنهای مرتبط با بیماریزایی، بویژه ژن PR-2 و ژنهای پاسخ دفاعی بویژه ژن HSR-203jمشاهده شد. بهرحال بیان ژنهای مذکور در نمونه های شاهد، تغییر پیدا نکرد. نتایج نشان داد که بین میزان بیان ژنها در نمونههای موردآزمایش و شاهد (گیاهان تیمارنشده با گلیفوسیت) ارتباط معنیداری وجود دارد. نتایج نشان داد که تیمار با گلیفوسیت، می تواند با القای پروتئینها و ژنهای پاسخ دفاعی، نوعی مقاومت اکتسابی سیستمی نسبت به باکتریهای بیماریزای گیاهی، ایجاد کند.

تجزیه و تحلیل جدایههای Pyricularia oryzae به دست آمده از برنج و علف های هرز با نشانگر rep-PCR و آزمون بیماریزایی
قارچ Pyricularia oryzae عامل بیماری بلاست و لکه برگی روی برنج و بیش از 50 گونه گیاهی در خانواده Poaceae از جمله محصولات زراعی و علفهای هرز میباشد. برای این پژوهش، نمونهبرداری در سال 1396 از مزارع برنج در استانهای گیلان و مازندران انجام شد. بررسیهای ریخت شناسی نشان داد جدایههای به دست آمده از میزبانهای برنج، چسبک، سوروف، سورگوم وحشی و پاسپالوم متعلق به گونه P. oryzae میباشند. نتایج آزمون بیماریزایی نشان داد جدایههای به دست آمده از هر میزبان روی همان میزبان قدرت بیماریزایی بالاتری دارند و روی میزبانهای دیگر یا بیماریزا نبوده، و یا قدرت بیماریزایی کمتری دارند. تجزیه و تحلیل خوشهای دادههای حاصل از انگشتنگاری DNA با استفاده از تکنیک مولکولی rep-PCR نشان داد که جدایههای به دست آمده از میزبانهای مختلف در سه دودمان کلونی قرار میگیرند. به طور کلی، در هر سه دودمان کلونی شناسایی شده، جدایههایی از میزبانهای مختلف وجود داشت و ارتباطی بین الگوی باندی با منطقه جمعآوری آنها دیده نشد.

اطلاعات جدید پیرامون شبه قارچ های مولد زنگ سفید گیاهان در ایران
بیمارگرهای عامل بیماری زنگ سفید گیاهان (Albuginales, Oomycota) شامل سه جنس Albugo، Pustula و Wilsoniana هستند که روی میزبانهای تیره Brassicaceae، راسته Caryophyllales و زیر رده Asteridae بیماریزا می باشند. این تحقیق با هدف ارائه اطلاعات جدید از راسته Albuginales در ایران انجام شد. در راستای نمونه برداری از زنگ سفید گیاهان، بازدیدهایی از برخی مناطق شمالی و شرقی ایران در بازه زمانی 1396-1399 صورت گرفت. بر اساس صفات ریخت شناسی و داده های مولکولی مبتنی بر ژنهای ITS و Cox2 گونههای Albugo lepidii روی Lepidium sativum ، A. koreana روی Camelina transcaspica، Albugo arenosa از Strigosella africana، A. candida روی میزبان های متعدد، A. occidentalis از اسفناج و Wilsoniana portulacae روی Portulaca sp. شناسائی شدند. بر اساس نتایج حاصل از این تحقیق، وقوع گونه A. candida روی میزبان های Eruca sativa ،Raphanus sativus و Goldbachia laevigata از ایران و روی میزبانهای Sinapis arvensis ،Savignya parviflora ،Isatis leuconeura و Sisymbrium altissimum برای دنیا بر اساس ریختشناسی و دادههای مولکولی تایید می شود، گونه Sisymbrium septulatum نیز به عنوان میزبان جدید برای آن گزارش میشود. همچنین بیماری زنگ سفید با عامل A. candida به عنوان اولین بیماری روی جنس Savignya از ایران گزارش می شود. آرایه های Albugo lepidii و A. koreana برای میکوبیوتای ایران جدید می باشند. توصیف کامل همراه با تصاویر و موقعیت فیلوژنتیکی برای دو گونه Wilsoniana portulacaeو Albugo occidentalis ارائه می گردد.

تأثیر گل گوگرد و چند کود شیمیایی بر فعالیت نماتد سیستی چغندرقند (Heterodera schachtii) در شرایط میکروپلات
کاهش خسارت نماتد سیستی چغندرقند Heterodera schachtii یکی از زمینههای تحقیقاتی کشورهای تولیدکننده چغندرقند است. کودهای شیمیایی علاوه بر افزایش حاصلخیزی میتوانند خسارات ناشی از نماتدهای انگل گیاهی را کاهش دهند. در این پژوهش تأثیر سولفاتهای آمونیوم، کلسیم، روی و پتاسیم، نیترات پتاسیم، سیلیکات سدیم، سوپرفسفاتتریپل، سکوسترین آهن و گوگرد بر فعالیت H. schachtii و خصوصیات رشدی چغندرقند رقم حساس شریف در دو آزمایش مطالعه شد. آزمایش اول در گلدانهای پلاستیکی حاوی شش کیلوگرم خاک زراعی و ماسه رودخانهای (1:1) سترون و آزمایش دوم در گلدانهای حاوی نه کیلوگرم خاک زراعی غیرسترون، بهترتیب به مدت چهار و پنج ماه، در شرایط میکروپلات انجام شد. کودها قبل از کشت با خاک گلدانها مخلوط و مایهزنی نماتد با اضافه کردن 200 گرم خاک آلوده حاوی 18 سیست پر با 4200 تخم انجام شد. نتایج نشان داد که کودهای شیمیایی و گوگرد تأثیر معنیداری بر رشد چغندرقند آلوده در خاک مخلوط سترون و خاک زراعی نداشته و بعضاً باعث کاهش آن شدند. در خاک مخلوط سترون، سولفات روی 50 کیلوگرم، سکوسترین آهن پنج کیلوگرم، سیلیکات سدیم 10 کیلوگرم و سوپرفسفاتتریپل 100 کیلوگرم در هکتار باعث کاهش نسبی بیش از 67% جمعیت نهایی و فاکتور تولیدمثل نماتد گردیدند. در خاک زراعی، این شاخصها در تیمارهای سوپرفسفاتتریپل 100 کیلوگرم و گوگرد 100 و 200 کیلوگرم/هکتار کمتر از سایر تیمارها افزایش نشان دادند، ولی با شاهد در یک سطح آماری قرار گرفتند. آزمایش سوم به منظور بررسی تأثیر تیمارها بر تفریخ تخم نماتد درون ظروف کوچک حاوی خاک آلوده در گلخانه صورت گرفت. نتایج نشان داد که بیشترین تفریخ مربوط به تیمار سولفات آمونیوم 200 کیلوگرم/هکتار بود که از نظر آماری با سوپرفسفاتتریپل 100 کیلوگرم/هکتار در یک سطح آماری قرار گرفت.

بررسی فاکتورهای بیماریزایی قارچ Puccinia graminis f. sp. tritici عامل بیماری زنگ سیاه گندم در استان اردبیل در سه سال متوالی (1396-1398)
زنگ سیاه یا ساقه گندم با عامل قارچی (Pgt) Puccinia graminis f. sp. tritici یکی از شایعترین و خسارتزاترین بیماری-های گندم در جهان و ایران بهشمار میآید. در شرایط مساعد و تشکیل شدن هرم بیماری، بیمارگر توانایی از بین بردن محصول گندم در مدت زمان کوتاه را دارد. قارچ Pgt بهدلیل تکمیل نمودن چرخه جنسی، جهش و هیبریداسیون از تغییر پذیری ژنتیکی بالایی در ساختار جمیعت برخوردار میباشد و هم این امر سبب ایجاد نژادهای جدید (تنوع نژادی) با الگوی بیماریزایی متفاوت میشود. یکی از نژادهای پرآزار و مهم قارچ Pgt که در دهههای اخیر ظاهر شده و خسارتهای فراوانی به محصول گندم در سطح جهان تحمیل نموده است، نژاد (Ug99) TTKSK میباشد. این نژاد در داخل کشور نیز شناسایی و گزارش شده است. با این حال مطمئنترین، سالمترین و مقرون به صرفهترین روش مدیریت و کنترل این بیماری، استفاده از مقاومت ژنتیکی است. آشنایی دقیق از ساختار ژنتیکی جمعیت بیمارگر (نژادها) و آگاهی از فاکتورهای (ژنهای) بیماریزایی نژادهای Pgt در هر منطقه اولین گام و به مشابه نقشه راه برای رسیدن به مقاومتهای موثر و پایدار ژنتیکی می-باشد. یکی از راههای بررسی روند تغییر الگوی بیماریزایی نژادها در هر منطقه با رصد فاکتورهای بیماریزایی روی ژنوتیپهای افتراقی زنگ سیاه در خزانههای تله میسر میگردد. بدین منظور در سه سال متوالی (1398-1396) پایش فاکتورهای بیماریزایی نژادهای Pgt و همچنین اثر بخشی ژنهای مقاومت موجود در ژنوتیپهای افتراقی بینالمللی نسبت به نژادهای Pgt موجود در استان اردبیل، بررسی شدند.

شناسایی بیوانفورماتیک دو ویروس جدید از پسته ایرانی
پسته یکی از گیاهان پر اهمیت اقتصادی بوده و به خانواده Anacardiaceae تعلق دارد. گزارشهای مرتبط با عوامل ویروسی در این گیاه به ویروئید کوتولگی رازک، آمپلوویروس آ پسته و ویروس ب پسته محدود است. در مطالعه حاضر به منظور بررسی حضور احتمالی ویروسها در پسته ایرانی به واکاوی دادههای ترنسکریپتومیک عمومی پرداخته شد. دوازده داده با خوانش جفت شده از پایگاه داده SRA قابل دسترس در وبسایت NCBI اخذ گردید. آراناِهای گیاهی پس از همردیف سازی دادهها بر روی ژنوم مرجع پسته به وسیله نرمافزار Hisat2، حذف گردید. توالیهای غیر گیاهی به وسیله نرم افزارهای MEGAHIT و Trinity مونتاژ و نتیجۀ حاصله به منظور شناسایی توالیهای ویروسی توسط نرمافزار BLASTx در توالیهای مرجع ویروسی مورد جستجو قرار گرفت. دو ویروس ناشناس متعلق به خانواده Kitaviridae شناسایی و موقتاً ویروس ایکس پسته (MT334618-20) و ویروس وای پسته (MT362605-06) نام گذاری گردیدند. بر اساس ساختار ژنوم، تشابه ژنتیکی و روابط فیلوژنتیک پیشنهاد میشود این دو ویروس ناشناخته به ترتیب به عنوان عضوی از جنسهای Higrevirus و Cilevirus در نظر گرفته شوند. تعداد بالای توالیهای ویروسی در نمونههای ریشه و برگ، احتمال آلودگی سیستمیک ویروسی ایکس پسته را مطرح مینماید. براساس بررسیهای صورت گرفته، این اولین گزارش از توالی ژنومی عامل ویروسی احتمالی بر روی پسته ایرانی میباشد.
