تأثیر دما، مرحله رشدی و زه‏آب گیاهان میزبان و غیرمیزبان روی میزان تفریخ تخم نماتد سیستی کلم (Heterodera cruciferae)

چکیده

اثر دما، مرحله رشدی گیاه میزبان و زه‏آب گیاهان میزبان (کلم قمری و کلم برگ) و غیرمیزبان (کاهو) در مراحل رشدی مختلف و در شرایط دمایی مختلف شامل 5، 10، 15، 20 و 25 درجه سانتی‏گراد بر تفریخ تخم نماتد سیستی کلم (Heterodera cruciferae) مورد بررسی قرارگرفت. زه‏آب گیاه غیرمیزبان و گیاهان میزبان با دوره رشدی سه و شش ماهه در چهار مرحله رشدی (دو، چهار، شش و هشت برگی) جمع‏آوری شدند. غلظت‏های مختلف زه‏آب و رقت‏های یک چهارم و یک شانزدهم آن تهیه و همراه با دو شاهد منفی (زه‏آب گلدان‏های تیمارنشده و آب) و یک شاهد مثبت (محلول 3/0 مولار کلریدروی) مورد استفاده قرار گرفتند. تعداد 25 سیست نماتد در داخل این محلول‌ها در ظروف پلاستیکی 24 چاهکی قرار گرفته و با فواصل یک هفته، تعداد لاروهای تفریخ شده تا سه هفته یادداشت‏برداری گردید. نتایج نشان داد که رقت یک چهارم زه‏آب گیاه کلم برگ با دوره رشدی شش‏ماهه در مرحله رشدی شش برگی در دمای °C20، موجب افزایش میزان تفریخ تخم گردید.

چکیده [English]

Stimulation of egg hatching of cabbage cyst nematode (Heterodera cruciferae) were investigated by soil drainages of host (kohlrabi and white cabbage) and non-host (lettuce) plants in different life stages, under different temperature conditions including 5, 10, 15, 20 and 25°C. Soil drainages of non-host plant and three and six-months-old seedlings of host plants were collected at 2, 4, 6 and 8 leaves developmental stages. Different concentrations of soil drainagesincluding undiluted soil drainages, soil drainagesdiluted 1:4 or 1:16 with water were used. Pot’s draining water and tap water were used as two negative controls and 0.3 M ZnCl2 solution as a positive control. Twenty-five cysts were stored in a 24-well plastic plate containing the soil drainagessolutions. The rate of eggs hatching was noted with a week intervals up to three weeks. The results showed that dilution of six-months-old white cabbage soil drainages1:4 at six leaves stage enhanced stimulation of hatching at 20°C compared with other treatments.
